RESUMO

Cadmium (Cd) is a widespread and highly toxic environmental pollutant, seriously threatening animal and plant growth. Therefore, monitoring and employing robust tools to enrich and remove Cd from the environment is a major challenge. In this work, by conjugating a fluorescent indicator (CCP) with a functionalized glass slide, a special composite material (CCPB) was constructed to enrich, remove, and monitor Cd2+ in water rapidly. Then Cd2+ could be effectively eluted by immersing the Cd-enriched CCPB in an ethylenediaminetetraacetic acid (EDTA) solution. With this, the CCPB was continuously reused. Its recovery of Cd2+was above and below 100 % after multiple uses by flame atomic absorption spectrometry (FAAS), which was excellent for practical use in enriching and removing Cd2+ in real aqueous samples. Therefore, CCPB is an ideal material for monitoring, enriching, and removing Cd2+ in wastewater, providing a robust tool for future practical applications of Cd enrichment and removal in the environment.

Cadmium (Cd) contamination of farmland soils is a growing concern because of its highly toxic impact on ecosystems and human health. Chelator-assisted washing and chemical immobilization are effective remediation strategies for Cd-contaminated soils. Ethylenediaminetetraacetic acid (EDTA) has traditionally been used for soil washing, but its persistence in the environment and subsequent toxicity have raised significant ecological concerns. Consequently, biodegradable chelators have gained increasing attention as eco-friendly alternatives to the persistent chelator, EDTA. Therefore, this study evaluated the performance and efficacy of three biodegradable chelators: L-glutamate-N,N'-diacetic acid (GLDA), methylglycine-diacetic acid (MGDA), and 3-hydroxy-2,2'-iminodisuccinic acid (HIDS) in comparison to EDTA for remediating a real Cd-contaminated agricultural soil. The influence of treatment parameters, including chelator variants, washing time, chelator concentration, solution pH, and liquid-to-soil ratio (L/S) on Cd extraction was studied and optimized to attain the maximum removal rate. Following chelator-assisted washing, the efficacy of a stabilization preference combining FeCl3 and CaO in reducing the leaching potential of residual Cd in chelator-washed soil residues was also investigated. GLDA demonstrated comparable Cd extraction efficiency to EDTA, and the Cd extraction efficiency was found to be positively correlated with the soil washing parameters. However, under the optimized conditions (chelator concentration: 10 mmol L-1; washing time: 3 h; solution pH: 3; L/S ratio: 10:1), GLDA exhibited a higher Cd extraction rate than EDTA or the other chelators. Furthermore, a post-treatment process incorporating FeCl3 and CaO substantially diminished the water-leachable Cd content in the resultant soil residues. The proposed remediation strategy, which combines chemically assisted washing and stabilization, could be a practical option for extracting bulk Cd from soil and reducing the leaching potential of residual Cd.

Streck tubes are commonly used to collect blood samples to preserve cell-free circulating DNA. They contain imidazolidinyl urea as a formaldehyde-releasing agent to stabilize cells. We investigated whether the released formaldehyde leads to crosslinking of intracellular proteins. Therefore, we employed a shotgun proteomics experiment on human peripheral blood mononuclear cells (PBMCs) that were isolated from blood collected in Streck tubes, EDTA tubes, EDTA tubes containing formaldehyde, or EDTA tubes containing allantoin. The identified crosslinks were validated in parallel reaction monitoring LC/MS experiments. In total, we identified and validated 45 formaldehyde crosslinks in PBMCs from Streck tubes, which were also found in PBMCs from formaldehyde-treated blood, but not in EDTA- or allantoin-treated samples. Most were derived from cytoskeletal proteins and histones, indicating the ability of Streck tubes to fix cells. In addition, we confirm a previous observation that formaldehyde crosslinking of proteins induces a +24 Da mass shift more frequently than a +12 Da shift. The crosslinking capacity of Streck tubes needs to be considered when selecting blood-collection tubes for mass-spectrometry-based proteomics or metabolomic experiments.

This study describes the development of two highly sensitive immunosensor platforms for the trace determination of copper ions, Cu(II), in drinking water. These platforms were a microwell-based enzyme-linked immunosorbent assay (ELISA) and a kinetic exclusion assay (KinExA) with a KinExATM 3200 immunosensor. Both ELISA and KinExA were developed utilizing the same antibody and coating reagent. The antibody was a mouse monoclonal antibody, designated as 8D66, that specifically recognized Cu(II)-ethylenediamine tetraacetic acid complex (Cu(II)-EDTA) but did not recognize Cu(II)-free EDTA. The 8D66 monoclonal antibody was generated by the fusion of spleen cells of an immunized BALB/c mouse with SP2/0-Ag14 myeloma cells. The immunogen was a protein conjugate of Cu(II)-EDTA with keyhole limpet hemocyanin protein. The coating reagent was Cu(II)-EDTA covalently linked to bovine serum albumin protein (Cu(II)-EDTA-BSA). Both assays involved the competitive binding reaction between Cu(II)-EDTA complexes, formed in the sample solution, and Cu(II)-EDTA-BSA conjugate which has been immobilized onto ELISA plates (in ELISA) or polymethylmethacrylate beads (in KinExA) for a limited quantity of binding sites of the 8D66 antibody. In ELISA, color signals were generated by a peroxidase-labeled secondary antibody and 3,3',5,5'-tetramethylbenzidine substrate. In KinExA, a fluorescein isothiocyanate-labeled secondary antibody was used to generate KinExAgram (trend-line fluorescence responses vs. time). The conditions of both ELISA and KinExA were investigated, and the optimum procedures were established. Both ELISA and KinExA were validated, and all validation parameters were acceptable. Many different metal ions that are commonly encountered in drinking water did not interfere with the Cu(II) analysis by both ELISA and KinExA. Both assays were applied to the determination of Cu(II) in drinking water with satisfactory accuracy and precision. Both assays were compared favorably with inductively coupled plasma atomic emission spectroscopy in terms of their abilities to accurately and precisely determine Cu(II) in drinking water samples. A comparative evaluation of ELISA and KinExA revealed that KinExA had a higher sensitivity and better precision than ELISA, whereas both assays had comparable accuracy. Both ELISA and KinExA were superior to the existing atomic spectrometric methods for Cu(II) in terms of sensitivity, convenience, and analysis throughputs. The proposed ELISA and KinExA are anticipated to effectively contribute to assessing Cu(II) concentrations and control the exposure of humans to its potential toxicities.

Objetivo: Este estudo teve como objetivo avaliar a resistência de união do cimento Biodentine® à dentina radicular após a utilização de diferentes irrigantes finais. Método: Vinte dentes humanos extraídos tiveram seu terço médio radicular cortado em fatias que foram submersas em hipoclorito de sódio 2,5% e posteriormente divididas aleatoriamente em 4 grupos experimentais (n=15) conforme o irrigante final utilizado (1) água destilada (controle), (2) QMixTM, (3) ácido cítrico 10%, (4) EDTA 17%. Após a imersão na substância teste as amostras foram preenchidas com o cimento Biodentine e imersas em solução salina tamponada com fosfato (PBS) por um período de 7 dias. O teste de push out foi realizado e os valores de resistência de união em Mpa foram obtidos. Os dados foram analisados pelos testes de Kruskal Wallis e Studend- Newman-Keuls. Resultados: Os piores valores de união foram obtidos após a utilização do EDTA enquanto a água destilada, o QMix e o ácido cítrico apresentaram resultados estatisticamente semelhantes entre si. Conclusão: A remoção da smear layer não resultou em melhora nos resultados de união do cimento Biodentine.(AU)


Objective: This study aimed to evaluate the bond strength of Biodentine® cement to root dentin after the use of different final irrigants. Method: Twenty extracted human teeth had their middle root third cut into slices that were submerged in 2.5% sodium hypochlorite and then randomly divided into 4 experimental groups (n=15) according to the final irrigant used (1) distilled water (control), (2) QMixTM, (3) 10% citric acid, (4) 17% EDTA. After immersion in the test substance the samples were filled with Biodentine cement and immersed in phosphate buffered saline (PBS) for a period of 7 days. The push out test was performed and the bond strength values in MPa were obtained. The data were analyzed by Kruskal Wallis and Studend- Newman-Keuls tests. Results: The worst bond values were obtained after using EDTA while distilled water, QMix and citric acid showed statistically similar results to each other. Conclusion: Removal of the smear layer did not result in improved bonding results of Biodentine cement.(AU)

Ni is often present in plating wastewater as a complexing state. It is difficult to remove this Ni using traditional chemical precipitation technology. In this study, a bipolar membrane electrodialysis system was used to recover Ni in the form of Ni(OH)2 from plating wastewater containing Ni-ethylenediaminetetraacetic acid (Ni-EDTA) without adding chemical reagents. The stable structure of Ni-EDTA can be destroyed by H+ produced by the bipolar membrane to obtain free Ni2+, which can combine with OH- produced by the bipolar membrane to form Ni(OH)2. When the electrolyte Na2SO4 concentration, current density and initial Ni-EDTA concentration were 0.2 mol/L, 16 mA/cm2 and 1000 mg/L, respectively, 99.0% of Ni-EDTA was removed after 32 h. When the system was used to treat actual plating wastewater, 92.1% of Ni-EDTA was removed and 88.7% was recovered. When the number of wastewater compartments in the system was increased from one to three, the current efficiency increased from 1.7% to 5.8%, and the specific energy consumption decreased from 0.39 to 0.19 kW h/g. The results of an X-ray diffraction study indicate that the Ni(OH)2 obtained in this study is similar to commercial Ni(OH)2. Moreover, the recovery mechanism of Ni-EDTA was analysed. Thus, bipolar membrane electrodialysis can be regarded as an effective method to recover Ni from wastewater containing Ni-EDTA.

Ni-EDTA is widely present in electroplating effluents. It cannot be effectively removed by traditional wastewater treatment methods due to its chemical stability. In this study, copper sulphide/cuprous sulphide doped zero-valent iron@carbon (ZVI@C/CuS/Cu2S) was prepared to active peroxymonsulphate (PMS) to decomposition Ni-EDTA. The ZVI@C/CuS/Cu2S + PMS process shows excellent performance under neutral or even alkaline conditions. This is due to the acceleration of ZVI electron transport by CuS/Cu2S, the autocatalysis of CuS/Cu2S itself, and the synergistic effect of CuS/Cu2S and Ni-EDTA. The removal efficiency of 50 ppm Ni-EDTA electroplating effluents reached 99.53% at 10 min, and the discharge water can meet the Chinese emission standard. The influences of the main parameters such as initial pH value, catalyst, PMS and initial Ni-EDTA concentration on removal efficiency was systematically investigated.

ABSTRACT Objective: To assess the efficacy of 5% Glycolic Acid (GA), 17% ethylenediaminetetraacetic acid (EDTA) and 7% maleic acid (MA), in removing the smear layer (SL). Material and Methods: For the experiment, forty single-rooted human teeth were selected. To perform the chemo‑mechanical preparation, the root canals were instrumented to an apical size of #30, along with simultaneous irrigation of 2.5% NaOCl. The samples were allotted to the experimental groups based on the final irrigating solution (n=10): (1) The GA group: 05%, (2) the EDTA group: 17%, (3) the MA group: 7%, and (4) the control group: Distilled water. The teeth were evaluated for the presence or absence of SL using SEM. Results: Comparing the 5% GA, 7% MA and 17% EDTA groups, no statistically significant differences were found at the coronal and middle thirds (p>0.05). However, in the apical third, MA had greater efficacy than EDTA (p=0.002) and GA (p=0.041), with a significant difference in the SL removal, while there was no significant difference between the latter (p=0.148). Conclusion: Thus, it was concluded that 7% maleic acid as a final irrigating solution is more efficacious than 17% EDTA and 5% glycolic acid in eliminating the smear layer from the apical portion of the root canal.

In this study, we tested the ability of a solution of effective microorganisms (EM) to remove cadmium from soil. Experimental results revealed that EM had an overall cadmium removal efficiency of 90.5% after 7 days of electrokinetic (EK) treatment. During EK treatment, EM exhibited a low initial pH of 3.6 and a high conductivity of 7.0 mS/m; therefore, they reduced the pH of the anode after an electric field was applied. EM had a surface tension of 50.3 dyne/cm and exhibited biosurfactant property in the EK experiments. The cadmium removal efficiency of EM in soil was compared with that of tap water, citric acid, and ethylenediaminetetraacetic acid (EDTA). The results revealed that after 7 days of EK treatment, EM had a higher cadmium removal efficiency than did citric acid (72.3%), EDTA (75.4%), and tap water (21.7%). This result can be partly attributed to the biosurfactant property of EM, which enables them to penetrate deeply into the soil matrix and thus dissolve a high quantity of pollutants. Overall, the results of this study indicate that EM can serve as an economic and efficient biosurfactant for removing cadmium from soil in EK applications.

While iron overload disorder (IOD) and related disease states are not considered a common occurrence in domestic equids, these issues appear prevalent in black rhinoceroses under human care. In addressing IOD in black rhinos, altering dietary iron absorption and excretion may be the most globally practical approach. A main option for treatment used across other species such as humans, is chelation therapy using iron-specific synthetic compounds. As horses may serve as an appropriate digestive model for the endangered rhinoceros, we evaluated the potential use of the oral iron chelator N,N-bis(2-hydroxybenzyl)ethylenediamine-N,N-diacetic acid (HBED) in horses for safety and efficacy prior to testing in black rhinoceros. Health and iron digestibility and dynamics were assessed in horses (n = 6) before, and after treatment with HBED (50 mg/kg body weight) for 8 days using a crossover design with serum, faecal and urine collection. A preliminary pharmacokinetic trial was also performed but no trace of HBED was found in serially sampled plasma through 8 h post-oral dosing. HBED increased urinary iron output in horses compared to control by 0.7% of total iron intake (p < 0.01), for an average of 27 mg urinary iron/day, similar to human chelation goals. Blood chemistry, blood cell counts and overall wellness were not affected by treatment. As healthy horses are able to regulate iron absorption, the lack of change in iron balance is unsurprising. Short-term HBED administration appeared to be safely tolerated by horses, therefore it was anticipated it would also be safe to administer to black rhinos for the management of iron overload.

Soil enzyme activities and microbial communities have a good response to the remediation effect of heavy metal-contaminated soils. To evaluate the effect of three commonly used washing agents, ferric chloride (FC), ethylenediaminetetraacetic acid (EDTA) and ethylenediamine-tetra-methylenephosphonic acid (EDTMP) on soil enzyme activities and microbial community in cadmium (Cd)-contaminated agricultural soil were collected from farmland near a non-ferrous metal smelter. The soil enzyme activities, microbial community, chemical forms of Cd and some physicochemical properties of the soil washed with different washing solutions were determined. The results showed that the three washing solutions had moderate removal efficiencies for Cd in the tested soil and the breakdown product of EDTMP has a certain stabilizing effect on Cd. The geometric mean and the integrated total enzyme activity index showed that soil washing with FC and EDTA was more beneficial to the restoration of biochemical functions than that with EDTMP. After soil washing, the Chao1 index of bacteria increased, and the microbial community structure changed. Pearson correlation analysis and redundancy analysis (RDA) indicated that the three washing solutions affected soil enzyme activities and microbial community by altering soil nutrient, total Cd concentration and Cd fractions in soils.

Because of the essential roles of SARS-CoV-2 papain-like protease (PLpro) in the viral polyprotein processing and suppression of host immune responses, it is a crucial target for drug discovery against COVID-19. To develop robust biochemical methodologies for inhibitor screening against PLpro, extensive characterization of recombinant protein is important. Here we report cloning, expression, and purification of the recombinant SARS-CoV-2 PLpro, and explore various parameters affecting its stability and the catalytic activity. We also report the optimum conditions which should be used for high-throughput inhibitor screening using a fluorogenic tetrapeptide substrate.

Since long-chain fatty acids work as the primary energy source for the myocardium, radiolabeled long-chain fatty acids play an important role as imaging agents to diagnose metabolic heart dysfunction and heart diseases. With the aim of developing radiogallium-labeled fatty acids, herein four fatty acid-based tracers, [67Ga]Ga-HBED-CC-PDA, [67Ga]Ga-HBED-CC-MHDA, [67Ga]Ga-DOTA-PDA, and [67Ga]Ga-DOTA-MHDA, which are [67Ga]Ga-HBED-CC and [67Ga]Ga-DOTA conjugated with pentadecanoic acid (PDA) and 3-methylhexadecanoic acid (MHDA), were synthesized, and their potential for myocardial metabolic imaging was evaluated. Those tracers were found to be chemically stable in 0.1 M phosphate buffered saline. Initial [67Ga]Ga-HBED-CC-PDA, [67Ga]Ga-HBED-CC-MHDA, [67Ga]Ga-DOTA-PDA, and [67Ga]Ga-DOTA-MHDA uptakes in the heart at 0.5 min postinjection were 5.01 ± 0.30%ID/g, 5.74 ± 1.02%ID/g, 5.67 ± 0.22%ID/g, and 5.29 ± 0.10%ID/g, respectively. These values were significantly lower than that of [123I]BMIPP (21.36 ± 2.73%ID/g). For their clinical application as myocardial metabolic imaging agents, further structural modifications are required to increase their uptake in the heart.

Liver-specific contrast agents (CAs) can improve the Magnetic resonance imaging (MRI) detection of focal and diffuse liver lesions by increasing the lesion-to-liver contrast. A novel Mn(II) complex, Mn-BnO-TyrEDTA, with a lipophilic group-modified ethylenediaminetetraacetic acid (EDTA) structure as a ligand to regulate its behavior in vivo, is superior to Gd-EOB-DTPA in terms of a liver-specific MRI contrast agent. An MRI study on mice demonstrated that Mn-BnO-TyrEDTA can be rapidly taken up by hepatocytes with a combination of hepatobiliary and renal clearance pathways. Bromosulfophthalein (BSP) inhibition imaging, biodistribution, and cellular uptake studies confirmed that the mechanism of hepatic targeting of Mn-BnO-TyrEDTA is the hepatic uptake of the amphiphilic anion contrast agent mediated by organic anion transporting polypeptides (OATPs) expressed by functional hepatocytes.

PURPOSE OF THE REPORT: Prostate-specific membrane antigen (PSMA) is a member of superfamily of zinc-dependent exopeptidases that is robustly expressed in prostate cancer cells and nonprostatic solid tumor neovasculature including microvessels of thyroid tumors. Its expression in differentiated thyroid cancer (DTC) has been confirmed in many recent studies, but systematic studies exploring PSMA expression in patients with DTC with thyroglobulin elevation and negative iodine scintigraphy (TENIS) are lacking. The aim of the present study was to evaluate the role of PSMA scan in TENIS patients with DTC. METHODS: Nine consecutive patients with DTC with proven TENIS syndrome (6 men and 3 women with age range 29-68 years and mean age of 48 years) underwent 18F-FDG PET/CT as per the institution protocol. Thereafter, they were subjected to 68Ga-PSMA-HBED-CC PET/CT as per the institution protocol within a week of FDG PET imaging. Prostate-specific membrane antigen expression (SUVmax) in the lesions was compared with 18F-FDG PET and CT scan findings. RESULTS: In 5 of 9 patients with TENIS, the metastatic lesions showed PSMA expression. A total of 14 lesions were seen on the CT scan. Prostate-specific membrane antigen PET detected 9 of 14 lesions (64.28%) (SUVmax ranging from 10.1 to 45.67; median SUVmax of 16.31), whereas FDG PET was positive in 11 of 14 lesions (78.57%). The lesions that showed PSMA uptake was localized to bones (5 of 9) and lungs (4 of 9). Two lesions that were localized to iliac crest and acetabulum were missed on FDG PET but were seen on CT and PSMA PET scan. CONCLUSIONS: The results of this pilot study indicate that 68Ga-HBED-CC-PSMA PET/CT demonstrates PSMA expression in TENIS patients with lesions being localized to the bones and lungs. 68Ga-PSMA PET/CT could be useful for the identification of TENIS patients who might benefit from PSMA-targeted radionuclide therapy.

BACKGROUND: The aim of this work was to study the possible synergic effect between mixtures with iron leonardite humate (L/Fe3+ ) and synthetic chelates iron (Ch/Fe3+ : o,oEDDHA /Fe3+ or HBED/Fe3+ ), and to reevaluate the classical chelate shuttle-effect model. Different molar ratios of L/Fe3+ :Ch/Fe3+ , different doses, and different sampling times were used in hydroponic and soil experiments using soybean (Glycine max) as a model Strategy I crop in calcareous conditions. Ligand competition between the humate and chelating agents was also examined. RESULTS: Iron humate participates in the chelate shuttle mechanism, providing available Fe to the chelating agent and then to the plants, showing a slight synergic effect. After a few days, the contribution of the chelates to the Fe nutrition decreases substantially, but the contribution of the humates is maintained. CONCLUSIONS: The most efficient ratio was two parts of iron humates and one part of iron chelate. In particular, HBED/Fe3+ was the most suitable iron chelate because its lasting effect fits the iron humate long-term effect better. The soluble iron in soil increased and the shoot-to-root iron translocation improved due to a synergic effect by a shuttle effect exerted by iron chelate in the mixture. © 2021 Society of Chemical Industry.

The effects of water soluble antioxidant (ascorbic acid and EDTA), fat soluble antioxidant (α-tocopherol) and amphiphilic antioxidant (ascorbyl palmitate; AP) on the chemical physics and bioaccessibility of ß-carotene loaded nanoemulsions (CNE) were investigated. During accelerated storage at 45 °C for 15 days, AP showed the highest protective actions against particle size growth, color fading, lipid oxidation, and ß-carotene degradation in CNE (p < 0.05). CNE with AP was then subjected to treat with cold plasma (CP) induced reactive species system under various powers and contact times compared to control. AP was able to protect physical and oxidative stabilities of CNE as well as ß-carotene integrity. The highest in vitro lipid digestibility, bioaccessibility and ß-carotene stability were found in CNE with AP (p < 0.05). However, those properties were lowered after CP exposure. The results indicated that AP was a promising antioxidant in improving physical stability, oxidative stability, ß-carotene retention, and ß-carotene bioaccessibility of CNE.

A novel adsorbent Mn-Fe layered double hydroxides intercalated with ethylenediaminete-traacetic (EDTA@MF-LDHs) was synthesized by a low saturation coprecipitation method. The behavior and mechanism of As(III) removed by EDTA@MF-LDHs were investigated in detail in comparison with the carbonate intercalated Mn-Fe layered double hydroxides (CO3@MF-LDHs). The results showed that EDTA@MF-LDHs had a higher removal efficiency for As(III) than As(V) with a broader pH range than CO3@MF-LDH. The large adsorption capacity of EDTA@MF-LDHs is related to its large interlayer spacing and the high affinity of its surface hydroxyl groups. The maximum adsorption capacity for As(III) is 66.76 mg/g at pH 7. The FT-IR and XPS characterization indicated that the removal mechanism of the As(III) on EDTA@MF-LDHs include surface complexation, redox, and ion exchange.

Sorbus commixta is a valuable hardwood plant with a high economical value for its medicinal and ornamental qualities. The aim of this work was to investigate the effects of the iron (Fe) source and medium pH on the growth and development of S. commixta in vitro. The Fe sources used, including non-chelated iron sulfate (FeSO4), iron ethylenediaminetetraacetic acid (Fe-EDTA), and iron diethylenetriaminepentaacetic acid (Fe-DTPA), were supplemented to the Multipurpose medium with a final Fe concentration of 2.78 mg·L-1. The medium without any supplementary Fe was used as the control. The pH of the agar-solidified medium was adjusted to either 4.70, 5.70, or 6.70. The experiment was conducted in a culture room for six weeks with 25 °C day and night temperatures, and a 16-h photoperiod with a light intensity of 50 mmol·m-2·s-1 photosynthetic photon flux density (PPFD). Both the Fe source and pH affected the growth and development of the micropropagated plants in vitro. The leaves were greener in the pH 4.70 and 5.70 treatments. The tissue Fe content decreased with the increase of the medium pH. The leaf chlorophyll content was similar between plants treated with FeSO4 and those with Fe-EDTA. The numbers of the shoots and roots of plantlets treated with FeSO4 were 2.5 and 2 times greater than those of the control, respectively. The fresh and dry weights of the shoot and the root were the greatest for plants treated with Fe-EDTA combined with pH 5.70. The calcium, magnesium, and manganese contents in the plantlets increased in the pH 5.70 treatments regardless of the Fe source. Supplementary Fe decreased the activity of ferric chelate reductase. Overall, although the plantlets absorbed more Fe at pH 4.70, Fe-EDTA combined with pH 5.70 was found to be the best for the growth and development of S. commixta in vitro.
